Techniques: Flow Cytometry, Concentration Assay

Tumor myeloid-derived lymphatic endothelial cell progenitors (M-LECP) are a subset of M2-type macrophages. Tumor sections were costained with antibodies against lymphatic vessel endothelial hyaluronan receptor 1 (LYVE-1) and CD204 (A), CD209 (B), or CD163 (C), typically used to identify M2-type tumor-associated macrophages. Circular arrangement of M-LECP cells [LYVE-1 and CD204/CD209 dual-positive clusters, indicated by white dashed line (A and B)] is reminiscent of nascent formation of a lymphatic vessel. Spatial organization of M-LECPs costained with other macrophage markers was observed in multiple tumor samples. White boxed areas in images in the third panels are shown in higher magnification in the fourth panels. Nuclei in merged images are identified by Hoechst stain. Scale bar = 100 μm.
